New PCR tests for Crassulaceae viruses and necroviruses in ornamental crops

From now on, we can test for the presence or absence of Kalanchoe mosaic virus (KMV), Kalanchoe latent virus complex (KLV) and Sonchus yellow net virus (SYNV) in Crassulaceae. We can also test for the necroviruses Tobacco necrosis virus-A (TNV-A), Tobacco necrosis virus-D (TNV-D), Olive mild mottle virus (OMMV) and Olive latent virus-1 (OLV-1) in leaf material of ornamental crops. These tests are carried out using the rapid, reliable and sensitive real-time PCR method.

The real-time PCR is a technique to rapidly propagate DNA  

This is done using pathogen-specific building blocks. The increase in DNA can be digitally monitored at any time. This makes it possible to confirm faster and with more certainty whether the pathogen is actually present in (suspected) plant material.  

Crassulaceae viruses 

From now on, we will be testing for Kalanchoe viruses exclusively using a new PCR method. This method replaces the old ELISA tests. The results of both tests correspond perfectly with one another. 

Various viruses occur in crops such as Kalanchoe and Sedum, including Kalanchoe mosaic virus (KMV), Kalanchoe latent virus complex (KLV) and Sonchus yellow net virus (SYNV). Various carlaviruses can be found in Kalanchoe, which is why the term ‘KLV species complex’ is used. 

Crassulaceae viruses on plant material (KMV, KLV and SYNV) 

  • Sample: maximum 2, 10 or 25 leaf(pieces)   
  • Fee test: € 11, € 22,80 en € 59,70 resp. per virus 
  • Duration of test: 4 days 

Necroviruses 

Viruses within the alpha- and betanecrovirus groups are transmitted by Olpidium spp. The new PCR tests detect tobacco necrosis virus-A (TNV-A), tobacco necrosis virus-D (TNV-D), olive mild mottle virus (OMMV) and olive latent virus-1 (OLV-1). The latter virus can also occur in strawberry roots.  

Necroviruses on plant material (TNV-A, TNV-D, OMMV and OLV-1) 

  • Sample: maximum 2, 10 or 25 leaf(pieces)   
  • Fee test: € 11, € 22,80 en € 59,70 resp. per virus 
  • Duration of test: 4 days   

 Please note: Previously, the terms TNV1 and TNV2 were listed on the certificates. However, these are the names of the antisera used at the time, not of the viruses themselves. TNV1 detected the TNV-A virus, whilst TNV2 detected the TNV-D and OMMV viruses.
